RD is definetly lead. The attitude fits perfectly.
Now I'd say, attitude-wise, Rarity should be the singer because she's the biggest attention whore, although she could easily have the best singing voice out of the six anyway, IMO. I know Pinkie sings the most and Fluttershy's songs are said to be the most beautiful, but Rarity's voice actor utilizes the most melodic voice out of them all(She also seems to take care of most of the more difficult voice acting
LIKE THE ROYAL CANTERLOT VOICE!)
Something makes me link Applejack with Bass...she emits some sort of calm strength/coolness and "down-to-earth" fits to her anyway (IMG:[

style_emoticons/default/happy.gif).
The Keyboard has the most sophisticated/technical air, so it could be something for Twilight, on the other hand, it's the most deep/bautiful/sophisticated in an emotional sense which would make it VERY interesting for Fluttershy.
Pinkie could do anything, she sings all the time, plays a shitload of instruments comes up with most songs, so she is definetly, together with Twilight, the most creative/guiding asset to the group.
I'd picture her more in the BG, though, which leaves the drums for her alone, driving the song forward, waiting out until she can go nuts in a solo, while Twi gets the Rhythm Guitar (Balanced between guiding drive and fleshing-out of the song, not wanting to show-off too mcuh and still contributing as much as possible).
