Finally understand it now. Windows encodes any selected image into a highly compressed JPEG file before setting it as the desktop wallpaper. That explains why the PNG's transparent background wasn't properly displayed on the desktop. The solution was to open the original image with a web browser first, before setting it as the desktop wallpaper.
