The way that I would take it is that you are and always will be what you see yourself as - if someone transitions, it's not because they "used to be" the other gender, but because they want their gender to be evident to others as well as themselves.
Maybe it's a stretch to claim that's what Rogers was saying. But I can't really claim to know. Based on the time he is from, I'd not be surprised either way.
I am old enough to have watched Mr. Rogers when he was still doing shows when I was young. He stopped when I was in grade school.
It's clear that the lines were directed to children who weren't sure if they were going to magically change or if they'd have to change. In fact, in this interview, he said directly that, that "sometimes, children think that they might change. They might have to change after a while. And y'know, we laugh about that now, but that's because we had that concern when we were little." Also, prior to talking about the song, he was talking about kids who were scared of getting their hair cut and scared of falling down the toilet, which just hammers in the actual message he wants to get across; that this song was meant for small children to dispel irrational fears or concerns that they might have, or just to clear up things they're confused about. It wasn't for children who felt that they were in the wrong body.
Mr Rogers was inclusive to everyone, we all know very well what his stance on being trans would be; "I like you just the way you are."

He realized that children needed help learning that their bodies will behave in predictable ways. It's inaccurate to assume that this video is Mister Rogers "weighing in on the transgender debate." In fact, the fear Mister Rogers talks about here--kids fear that their bodies will behave in ways they don't want, sounds an awful lot like gender dysphoria in young trans kids. Except for trans kids, that fear has already happened--they are living in a body that doesn't match their mental map of themselves.
It's Mister Rogers' deep empathy, his concern with addressing kids' deep fears, that I think we should take away from this video.
I choose to read as much empathy into what he says as I can. Because that's the kind of person he seemed like to me.
I also suspect that even if it was a statement about transgender youth, he would have spoken differently had he had as much personal experience with transgender individuals as many have today. I do not think he was particularly stuck in his ways when it came to how he looked at other human beings.

Mr Rodgers statement though, is that you are based in physical reality and from that you should be assured that we can predict with a usually high degree of certainty certain things. The time of his show was during the start of the movement to say that gender is entirely a social construct. He is accepting of people ahead of his time in terms of race, gender and orientation, but a childhood psychology perspective acknowledges that children could believe things about themselves that were not true quite easily.
What we do know that 50% is hard genetics, 20% is epigentics, and 30% is environment/social. If suddenly loads of people are identifying as a opposite gender or a new gender, especially in a very small time span, it points to the 30% of which social influence is a component. Humans compared to many animals have a higher proportion of the environment influence being social.
If you cut of children's social media access (or exposure to people that are heavy users) propensity to self identify trans drops by 30-60%. Then we also are finding that our entire human population is being over exposed to endocrine disruptors. That could be a horrifying influence on the 20%. As I also mentioned many variants of the pill also affect women's endocrine systems and pregnancy, especially if used longer than the fine print, and many people are encouraged or approved to do so.

My parents were ultra progressives for a while. I was even in a far left feminist film as a little kid. If it had been trendy to castrate chemically and physically back then, I have no doubt they would have.

It was very mid. They had a different distributor in each state, and internationally, so there aren't many copies available because there was a legal dispute when they wanted to release it on dvd. Its not worth any money for anyone to pay anything or go to court.
However... because its a movie with all the key words anti-establishment, resistance, activism, anti-war, anti-legal system, radical feminism etc... I have gotten out of jury duty all my life. No one can get a copy now to see that I was just a kid. They just look it up and see the credit and the synopsis and drop me off and put me on the do not call back list.
Almost everyone else I know has had to do jury duty at some stage.
